There has no single way to stop Full Screen Interstitital ad in Opera Mini.

Opera Devs forced us to this ad.
Poor Opera.

@williamskane This can be caused by a corrupted Opera installation, so be sure to reinstall Opera and check if that solves the problem

Well you can try this vpn app called Hotspotsheild.
Opera mini can connect while this vpn is on.
Annoying Ads totally ruins user experience.

@danielzr I guess Opera simply can't do that. Anyway, try checking Android settings and see if you can allow Opera Mini to play media on the background.
