I just installed this client, and have a few questions:
How do I change the default font?
How do I have the signature added to replies as well as to new messages?

Thank you in advance for your help.

The signature is added to replies.
Are you talking text-only messages or HTML? The answer to the font question depends on the message format...

The signature is still there. It's after the quoted text as Opera is made for replying in line. See https://en.wikipedia.org/wiki/Posting_style#Interleaved_style.

"Alt + p -> advanced -> fonts". You want to set both "Email compose" and "Email Compose (HTML)".

Note that when replying in HTML format to an HTML message, the compose font used will depend on the text-insert location in the composed source behind the scenes (you can goto "All Messages/Drafts", right-click the message and choose "view all headers and message" to see the styles in the HTML). In other words, when you reply, the font will be whatever font was used in the message you're replying to. However, if you hit ctrl + home, then backspace a few times, then enter and then ctrl + home again, Opera will use the "Email Compose (HTML)" font you have to set by default. When compose a new message instead of replying, Opera will user "Email Compose (HTML)" by default.

Thanks for your help. Found the fonts option and made changes.

Now, I'm looking for where to add a signature. I've looked everywhere, and cannot find where to add signature text. I must have found it before, but now cannot locate it.

              Thanks for any help here.

Menu -> Mail accounts -> select the account -> edit -> switch to Outgoing tab -> Edit Signature".

If you're going to create an HTML signature and you want to add a picture to it with <img src="">, don't add the img tag in the signature editor or it'll crash Opera Mail. Instead, goto "Menu -> help -> About Opera Mail" and take note of the mail directory location. Then, close Opera Mail and edit the signatureN.txt file in the mail folder where N is the number for the account as seen in accounts.ini. Doing things that way will avoid the crash. If you're not inserting an image, no need to do all of that.
