Are you using others Ads/contents/trackers blocker extension?? Like Adblock+, Privacy Badger, or native Opera adblocker?
To make Ublock working properly you need to disable all the others blockers, also antivirus real-time protection could interferes with ublock origin.
Check the new UBlock option "Allow access to search results" is active in extensions settings.
I don't know why all these guys are complaining about uBlock Origin... Tbh I don't see any ads anywhere.. not a single ad in YT.. so again.. check your ublock origins "allow access to search result" is active, and also make sure you're not using others adblockers or tracker blocker, neither the native opera gx ad blocker, cause that can cause problems with uBlock Origin.

Check the new option and see if it works again, also you shouldn't be using other Ad/tracker blockers, even the native Opera ad blocker must be disabled to let ublock origin works properly.
I just want to say ..... thank you so freaking much!!! This is simply the best ads/tracker blocker I've ever tried. Bring the internet back to its golden times ; ) and even better!

Switching to OperaGX 
it has tons of custom themes, the url bar in dark themes is... Dark and OMG...it looks so much better in my opinion,
I'm so satisfied now.

Plus I would put a collapse button for URL bar next to the "back" button, and another for bookmarks and tabs, exactly as the extensions tab already has.
Like tabs islands concept that allows you to hide bookmarks/URL bar/Tabs separately, in 1 click(or keyboard shortcut) for each section.
It would be awesome.
You should find that option in User Interface settings tab.
But tbh I think is a pretty useless option, as mentioned above, it's the exact same thing as searching in the address bar, so why would you have two search bars next to each other that do the exact same thing? Plus the search bar is also in the speed-dial page.
The Speed-Dial page is used by most of the users to have quick access to the most used web pages and links without having to resort to bookmarks, and usually just so you don't have to write "youtube" "gmail" and other sites you visit very often in the URL bar.
Therefore I agree with you @wickerman18 , having to access it with a keyboard shortcut or enabling it on the sidebar settings is not very user-friendly. that's all.
I also hope they put the option to reactivate it,
especially for new users who may be disappointed by the absence of a home button and then go back to a previous version or worse switch to another browser.
But don't get me wrong, I'm loving Opera One and its new design,
as far as I'm concerned these are only very small details.
I've installed opera one some days ago and still never tried Aria yet, everytime it says that "there is an error, refresh the page or try again later"or something like that, but never worked once.
Probably they're fixing some bugs in it.
I agree with you, the missing Speed Dial icon is the first thing I've noticed when I've installed Opera One.
It would be better to put it back next to the refresh button.
However you can set a keyboard shortcut that fixes the problem, while we hopefully wait for an update.
I've set Alt+A to open the Speed Dial/Home page in shortcuts settings, and it's very useful and comfortable, cause it allows you to switch the current page to "Speed Dials/Home" page without the need to open a new tab/use the side bar.
