You can use RegisterFirefoxPortable http://www.winhelponline.com/blog/register-firefox-portable-with-default-programs-in-vista/ to make Opera's launcher.exe available as a default browser in Windows. Then, set the launcher.exe as the default browser in Default Programs/Apps in Windows. Note that for the "Set apps by default" list, the one you want will say "Mozilla Firefox, Portable Edition". You can change that by editing ApplicationName in "HKEY_LOCAL_MACHINE\SOFTWARE\Clients\StartMenuInternet\FirefoxPortable\Capabilities" in the registry if you want.

I tested it with the latest build of Windows 10 and it still works for me. Just ignore the part I previously posted about looking for "Mozilla Firefox, Portable Edition" entry in Default Apps. Something changed in Windows 10 where it now gets the name for the browser in default apps from launcher.exe somehow.
Instead, after you launch RegisterFirefoxPortable.exe, point it to the path of the portable Opera's launcher.exe and click "register", in Start button -> settings -> apps -> default apps -> web browser, there will be an "Opera Internet Browser" entry (an extra one if you have Opera installed normally too) that you can set as the default.
Test it out by double-left-clicking an html file on your desktop so that Opera launches. Then, goto the URL
opera://about
and take a look at the install path to make sure you got the right Opera. Not needed to do this test if you only have one Opera installed. Are you talking about the portable version of Opera from https://www.opera.com/download (or the standalone installation in the installer's options) or are you talking about Opera from https://portableapps.com/apps/internet/opera_portable?

There's also https://browserchooser2.com/ that you can play around with instead if you can't get things to work.
